Even though having someone like Green Goblin as Spider-Man 3's villain would be familiar and expected, Morlun would bring something entirely new and terrifying. Morlun has made multiple attempts on Spider-Man's life, all with the intent to devour him and take Peter's power as his own. This makes him a natural enemy of Spider-Man, who he is terrifyingly insistent on pursuing. Morlun is one of the Inheritors, a race who gain power by feeding on totems, people across the multiverse who have been granted animal-themed powers. Although he will almost certainly not be the final boss, Spot's portals would be sure to give Spider-Man plenty of trouble during his adventure. Between his instantly recognizable look and strong and useful power, he has little difficulty leaving an impression. Spot may not have the most intimidating reputation among Spider-Man villains, and he's not among the villains Peter fought before the Marvel's Spider-Man game, but he is more than worth including in a video game. He could provide transportation for other villains, or move things around the city for the sake of their plans. However, Spot's portals open opportunities for several interactions. Spot has historically been more of a henchman than a major villain in his own right, so he is more likely to be a recurring foe or midboss. A good design team could make his teleporting spots look incredibly cool in action, be it in a cutscene or a fight against the villain. Spot's impressive powers make the advantages of including him in a video game obvious.
